Drug intelligence / Profile preview
iso-[131I]SGMIB-HF-22 is a radio-halogenated single-domain antibody (VHH or nanobody) targeting Claudin-18.2 (CLDN18.2), developed as a therapeutic radiopharmaceutical for gastrointestinal malignancies, particularly gastric and pancreatic cancers. The agent consists of the CLDN18.2-specific VHH clone HF-22 conjugated to the therapeutic radionuclide iodine-131 (131I) via the prosthetic group iso-SGMIB (iso-succinimidyl 4-guanidinomethyl-3-iodobenzoate). This specific formulation is designed to reduce renal and gastric retention, which are common limitations of VHH-based radiopharmaceuticals. In preclinical models, iso-[131I]SGMIB-HF-22 demonstrated high tumor uptake, rapid renal and gastric clearance, and significant dose-dependent tumor growth suppression without apparent toxicity.
